دستگاه کروماتوگرافی مایع متداول ترین روش برای ترکیبات غیر فرار مانند ترکیبات یونی یا پلیمرها ترکیبات ناپایدار حرارتی مانند مواد منفجره و ترکیبات فعال و پرتحرک مانند مواد بیولوژیکی می‌باشد. برای ترکیبات فرار، کروماتوگرافی گازی روش مناسب تری است اما تقریبا 80 درصد ترکیبات آلی شناخته شده غیر فرارند و میتوان روش کروماتوگرافی مایع را برای آنها به کار برد.

کروماتوگرافی مایع روش نسبتا ملایمی است به این معنی که در صورتیکه بتوان نمونه را در یک حلال مناسب حل کرد بسیاری از جداسازی ها در درجه حرارت معمولی انجام میشوند. مقدار نمونه میتواند از نانوگرم تا گرم متفاوت باشد. در صورت برخورداری از یک آشکار مناسب، این مقادیر در HPLC به پیکو گرم نیز میرسند.
